The Zoloft should be administered once a day, morning or evening.

Zoloft capsules should be administered during the meal.

Initial treatment

Depression and OCD

The Zoloft treatment should be started at a dose of 50 mg/day.

Panic disorder, PTSD and social anxiety disorder

The treatment will be started at a dose of 25 mg/day. After a week, the dose will be increased to 50 mg once daily. This dosing regimen has been shown to reduce the frequency of early adverse characteristic of panic disorder.

Dosage adjustment

Depression, TOC, panic disorder, social anxiety disorder and PTSD

Patients do not meet a 50 mg dose, a dose increase is possible. Dose changes should be done by levels of 50 mg at intervals of at least a week, up to a maximum of 200 mg/day. Dose changes should not be carried out more than once a week, taking into account the elimination half-life of the Zoloft which is 24 hours.

The therapeutic effect can occur within 7 days. However, longer periods are usually required to obtain a therapeutic response, in particular for the clicks.

Maintenance

The dose in a long-term treatment must correspond to the minimum effective dose, dosage adjustments based on the individual therapeutic response.

Depression

A more long-term treatment may also be appropriate for the prevention of recurrence of major depressive episodes (EDM). In the majority of cases, the recommended dose for the prevention of recurrence of EDM is identical to that used during the current episode. Depressive patients must be treated on a sufficiently long period of at least 6 months to ensure the disappearance of symptoms.

Panic disorder and TOC

Continued treatment in panic disorder or the TOC must be reassessed regularly, as relapse prevention has not been demonstrated in these disorders.

Pediatric patients

Children and adolescents with obsessive compulsive disorder

Between 13 and 17: initial dose of 50 mg once a day.

Between 6 and 12 years: initial dose of 25 mg once a day. The dose may be increased up to 50 mg once a day after a week.

Insufficient response, a secondary increase of the dose is possible by levels of 50 mg over a period of several weeks if necessary. The maximum dose is 200 mg per day.

It must however take account of the generally lower weight of children compared to adults an increase beyond 50 mg dose. Dose changes should not be carried out at intervals of less than a week.

Efficiency is not demonstrated in major depressive disorder of the child.

Use in hepatic insufficiency

The use of Zoloft in patients with liver disease must be made with caution. Inadequate hepatic must receive lower or more widely spaced doses (see warnings and precautions of employment). A brutal interruption must be avoided. In the case of the Zoloft treatment, the dose should be gradually reduced over a period of at least one to two weeks, to reduce the risk of withdrawal reactions (see warnings and precautions of use and adverse effects). If intolerable symptoms occur following a decrease in the dose or upon discontinuation of treatment, a resumption of the previously prescribed dose may be considered.
